Bioactivity of Verbascoside

In Vitro Study:

As a competitive PKC Inhibitor of ATP, Verbascoside has an IC50 of 25 μ M。 Verbascoside showed kis of 22 and 28 relative to ATP and histone, respectively μ M。 Verbascoside has effective antitumor activity on L-1210 cells with IC50 of 13 μ M[1]。 Verbascoside (5,10) μ M) Inhibition of 2,4-dinitrochlorobenzene (DNCB) – induced T cell costimulatory factors CD86 and CD54, proinflammatory cytokines and NF in thk-1 cells κ B pathway activation [2].

In Vivo Studies:

Verbascoside (1%) reduced the incidence of overall scratching behavior and the severity of skin lesions in a mouse model of 2,4-dinitrochlorobenzene (DNCB) – induced atopic dermatitis (AD). Verbascoside can also block the pro-inflammatory cytokine TNF in DNCB induced skin lesions- α, Expression of IL-6 and IL-4 mRNA [2]. Verbascoside (50100 mg / kg, IP) did not change the cold abnormal pain caused by chronic compressive injury (CCI). Verbascoside(200 mg / kg, IP) reduced allergy to cold stimulated acetone on day 3. Verbascoside also significantly reduced the behavioral changes associated with neuropathy. In addition, Verbascoside reduced Bax and increased Bcl-2 on day 3 [3].

Cell Experiment:

lymphocytic mouse leukemia L1210 cells (ATCC, CCL 219) contained 10% fetal bovine serum, 4 mM glutamine, 100 U / ml penicillin, 100 μ In the 24 well cluster plate of Dulbecco modified Eagle medium, 104 cells per well were sparsely laid/ Ml streptomycin sulfate and Verbascoside (dissolved in DMSO). Growth was monitored by counting the number of cells in the Coulter counter after 2 days of incubation in a humid atmosphere (5% CO2 in air) at 37 ℃. The IC50 value was calculated based on the linear regression line established for each test compound [1].

Animal Experiment:

In order to induce atopic dermatitis (AD) – like symptoms, rats [2] used 2,4-dinitrochlorobenzene (DNCB). In short, the dorsal hair of mice was removed with electronic scissors 2 days before DNCB treatment. Will 200 μ L of 1% DNCB (in acetone: olive oil = 4:1) was applied to shaved back skin for sensitization. Repeated attacks were performed at the same site, 0.2% DNCB every 3 days for about 2 weeks. Mice were divided into 4 groups (n = 6 in each group): (1) vehicle treated control, (2) DNCB treated only, (3) 1% Verbascoside (acetone: olive oil 4:1) – treated only, and (4) DNCB + 1% Verbascoside treated group[2].
